Development and Validation of Liquid Chromatography- Tandem Mass Spectrometric Method for Assay of Zidovudine in Human Urine
Abstract
A sensitive and selective liquid chromatographic-tandem mass spectrometric (LC-MSMS)method was developed to determine zidovudine (ZDV) in human urine. After solidphaseextraction with SPE cartridge, the urine sample was analyzed on a C18 column(Symmetry 3.5 μm; 75×4.6 mm i.d) interfaced with a triple quadrupole tandem massspectrometer. Positive electrospray ionization was employed as the ionization source. Themobile phase consisted of water-acetonitrile (20:80, v/v).The method was linear over aconcentration range of 10–1000 ng mL-1. The lower limit of quantitation was 10 ng mL-1.The intra-day and inter-day relative standard deviation across three validation runs overthe entire concentration range was <9.5%. The accuracy determined at threeconcentrations (150, 350 and 750 ng mL-1 ZDV) was within ±0.73% in terms of relativeerrors.Keywords: Zidovudine, quantification, liquid chromatography-tandem massspectrometry, human urine, determination
