Box Behnken Design for Optimization of Operating Conditions for Methyl Violet Decolorization using Momordica Charantia Peroxidase

Methyl violet is a triphenylmethane dye which is recalcitrant and it is difficult to degrade it by conventional methods. Enzyme based remediation methods have gained importance as they are cost effective, highly specific and eco friendly. Methyl violet was degraded using crude peroxidase from bitter gourd (Momordica charantia). To optimize the decolorization conditions of methyl violet by crude Momordica charantia peroxidase, Response Surface Methodology (RSM) using Box Behnken (BB) design was applied. A quadratic model was obtained for dye decolorization through this design. The optimum values for temperature, dye concentration, incubation time and enzyme volume were found to be 400C, 100 mg/L, 60 mins and o.75 ml respectively. The predicted decolorization under the optimum conditions determined by Response Surface Methodology was in close agreement with the experimental results. The model was found to be significant and the correlation coefficient was found to be 0.9029. The results confirmed that RSM based on Box-Behnken design was a reliable method to optimize the operating conditions of dye decolorization.Publikováno
