Abstract

Heterocyclic nucleus 1,3,4-thiadiazole constitutes an important class of compounds for new drug development. From the last decade, good research is going on the 1,3,4-thiadiazole ring, as a result of which, many research groups has developed novel clinically useful compounds. These compounds had shown its importance as anti-inflammatory, analgesic, antimicrobial, antitumor, antifungal, antimycobacterial, anticonvulsant, antidiabetic, antiviral agents etc. So far, the modifications of the 1,3,4-thiadiazole ring has proven highly effective way to find medicinally useful compounds with improved potency and less toxicity. The present review highlights the pharmacological importance of thione substituted 1,3,4-thiadiazol derivatives. This review also discusses the structure-activity relationships of the most potent compounds.  It can act as an important tool for medicinal chemists to develop newer compounds possessing 1,3,4-thiadiazole moiety that could be better agents in terms of efficacy and safety
