Estimation of Genetic Diversity in Some Species of Meliaceae Family through RAPD Technique

Abstract


Random amplified polymorphic DNA (RAPD) analysis has proved to be useful in determining genetic relationship and variation in many plant species, result of RAPD analysis can be used to determine genetic distances and relatedness within or among species, which can then be utilized in construction of phytogenetic. RAPD analysis could also provide useful insights into the phylogenetic relationship. RAPD is a polymerase chain reaction (PCR), which is based on dominant molecular marker technique. This technique has been used for genetic DNA finger printing. The RAPD technique is informative and relatively simple molecular tool that is used to estimate genetic relatedness among genotype, identify cultivars and construct genetic linkage maps. Morphological characters are suspected to environmental change and may not always clearly distinguish or correctly cluster closely related taxa. Molecular markers are often used to classify phylogenetic relationship. Several molecular methods are widely applied in plant systematic including isozymes, RFLP, RAPD, AFLP, DFP techniques based on dispersed repetitive DNA such as ISSR and SSR or microsatellite and restriction site variation of DNA.
