Multiplex PCR Assay for Rapid Detection of Different Strains of Escherichia coli Associated with Fish and Fishery Products

Abstract


A multiplex PCR (MPCR) assay was developed for the detection of Escherichia coli using selected primers designated on the species- and strains-specific genes such as hlyA, LT1 and phoA. The internal regions amplified were having a product size of 165, 275, and 903 bp, respectively. This MPCR assay had a sensitivity of detecting four cells of E. coli within 10 h in artificially contaminated shrimp tissue homogenate. The MPCR assay was found to be specific for E. coli, as it yielded negative results with other tested bacterial pathogens such as Salmonella typhimurium and Vibrio cholerae. Therefore, this developed MPCR assay could be used for the detection of different strains of E. coli (EHEC, EPEC, ETEC and EAEC) from clinical and environmental samples.
