Thymoquinone Induces Epigenetic Modulation in ROS Dependent Manner
Abstract
Traditional drugs have the ability to treat broad range of diseases by modulating the signaling pathways of inflammatory diseases and cancer. Epigenetic modulation is also one of the mechanisms already established as a process of action. Histone deacetylase (HDAC) inhibition leads to changes in gene expression promoting cell death. Thymoquinone (TQ), a bioactive compound from Nigella sativa seeds has shown to have an antineoplastic effect in many cancers. Here we have investigated HDAC inhibiting potential of TQ. It was observed that (TQ) inhibits the cell viability of HeLa and HepG2 cell lines in concentration dependent manner. It is able to enhanced the total oxidative species in both the cell lines. TQ treatment also causes a loss in mitochondrial membrane potential. The analysis of protein expression of HDAC1 and has shown a decrease in the level of HDAC1 and increase in the p21expression that confirming its ability to induce epigenetic modulation.
